An improved method for transformation of lettuce by Agrobacterium tumefaciens with a gene that confers freezing resistance BABT
Pileggi,Marcos; Pereira,Albanin Aparecida Mielniczki; Silva,Joandrei dos Santos; Pileggi,Sônia Alvim Veiga; Verma,Desh Pal S..
An efficient method for constructing transgenic lettuce cultivars by Agrobacterium tumefaciens was described by Torres et al., 1993. In the present work, an improvement of the above procedure is described and applied to transform the cultivar Grand Rapids with a mutated P5CS gene. The major modifications were concerned with turning more practical the transformation and regeneration protocols. Also we tried to improve transformation steps by increasing injured area in explants and prolonging co-cultivation with Agrobacteria (in larger concentration). A more significant selective pressure was used against non-transformed plants and bacteria. In these work we were concerned to obtain T1 and T2 seeds. The P5CS gene codes for a delta¹-pyrroline-5-carboxylate...

Increase of stress resistance in Lactococcus lactis via a novel food-grade vector expressing a shsp gene from Streptococcus thermophilus BJM
Tian,Hongtao; Tan,Jianxin; Zhang,Lifang; Gu,Xinxi; Xu,Wentao; Guo,Xinghua; Luo,Yunbo.
The effects of the expression of a small heat shock protein (shsp) gene from Streptococcus thermophilus on stress resistance in Lactococcus lactis under different environmental stresses were investigated in this study. pMG36e-shsp, an expression vector, was first constructed by inserting a shsp open reading frame (ORF) cloned from S. thermophilus strain St-QC into pMG36e. Then, a food-grade expression vector, pMG-shsp, was generated by deleting the erythromycin resistance gene from pMG36e-shsp. The transformation rate of pMG-shsp was comparable to that of pMG36e-shsp when each of these two vectors was introduced into L. lactis. These results demonstrated that the shsp ORF could successfully used as a food-grade selection marker in both pMG-shsp and...

Rosmarinus officinalis L. increases Caenorhabditis elegans stress resistance and longevity in a DAF-16, HSF-1 and SKN-1-dependent manner BJMBR
Zamberlan,D.C.; Amaral,G.P.; Arantes,L.P.; Machado,M.L.; Mizdal,C.R.; Campos,M.M.A.; Soares,F.A.A..
Improving overall health and quality of life, preventing diseases and increasing life expectancy are key concerns in the field of public health. The search for antioxidants that can inhibit oxidative damage in cells has received a lot of attention. Rosmarinus officinalis L. represents an exceptionally rich source of bioactive compounds with pharmacological properties. In the present study, we explored the effects of the ethanolic extract of R. officinalis (eeRo) on stress resistance and longevity using the non-parasitic nematode Caenorhabditis elegans as a model. We report for the first time that eeRo increased resistance against oxidative and thermal stress and extended C. elegans longevity in an insulin/IGF signaling pathway-dependent manner. These data...

Tolerance to the air exposition test of Hoplias lacerdae larvae and juvenile during its initial development BABT
Luz,Ronald Kennedy; Portella,Maria Célia.
The establishment of evaluation parameters for larvae and juvenile quality assessment is highly desirable. The experiment evaluated the Stress Resistance Rate (Re) on Hoplias lacerdae larvae and juveniles. Larvae on the 13th, 16th, 19th, 23rd and 26th days of life were subjected to Re evaluation tests. Twenty animals were stoked in one liter beaker and kept at 27 to 28.5°C. The following treatments were applied: E3 -3 minutes; E5 - 5 minutes; E7 - 7 minutes; E10 - 10 minutes and E15 -15 minutes on drying paper. After 24 hours Re was evaluated. It was observed that Re values showed an increase from 13th to 19th day in all treatments indicating an increasing resistance related to larval development. High Re rates were observed during the whole experiment,...
